Abstract

Ziel des ersten Projekts dieser Arbeit war die Totalsynthese von Bakkenolid A. Der Schlüsselschritt hierfür war die Entwicklung einer regioselektiven Methode zur 3-Funktionalisierung von Tetronaten mittels Pentadienyl-Verbindungen. Bis zur Realisierung der Reaktion waren detaillierte Untersuchungen bezüglich deren Reaktionsmechanismus und der damit einhergehenden Regioisomere nötig. Durch die Synthese diverser Pentadien-Buildingblocks konnte letztlich der optimale Reaktionspartner für eine regioselektive Tsuji-Trost-Reaktion gefunden werden. Eine vollständige Synthese von Bakkenolid A scheiterte jedoch aufgrund unüberwindbarer, sterischer Hinderung bei der anschließenden Diels-Alder Reaktion. Im zweiten Teil dieser Arbeit wurde die Totalsynthese von Macrocidin A angestrebt. Ein zentraler Teil der Retrosynthese-Strategie beruhte auf der erfolgreichen 3-Acylierung von Tetramsäuren gemäß Yoshii et al. Da die Ausbeuten dieser Methode aufgrund des bis heute ungeklärten Mechanismuses sehr schwer vorherzusagen sind, wurden deren Möglichkeiten detailliert untersucht. Hierbei zeigte sich eine starke Abhängigkeit von der Kettenlänge und v.a. dem Verzweigungsgrad in α-Position der verwendeten Carbonsäure. Die so erhaltenen 3-Acyl-Tetramsäuren wurden in ihre partiell bzw. global entschützten Derivate überführt und somit eine Substanzbibliothek aufgebaut. Diese wurde bezüglich ihrer cytotoxischen bzw. antibakteriellen Wirksamkeit untersucht. Hierbei konnten einige Verbindungen mit IC50-Werten im einstelligen µM-Bereich identifiziert werden. Die Erstellung einer eindeutigen Struktur-Wirkungs-Beziehung gelang allerdings nicht. Der entscheidende nächste Schritt in Richtung Totalsynthese von Macrocidin A war der Ringschluss zum 17-gliedrigen Makrocyclus. Ursprünglich nur als Entschützung der Phenol-Funktion geplant, gelang durch Palladium-Katalyse der direkte Ringschluss eines Nor-Macrocidin A Precursors. Hierbei handelt es sich um eine bis dato unbekannte, mit einer Williamson-Ether-Synthese vergleichbaren Methode zur Synthese von Makrocyclen.

A crucial objective of the work presented herein was the total synthesis of bakkenolid-A. To this end, a regioselective 3-functionalization of a tetronate using a pentadiene was established. For the refinement of the reaction extensive experiments concerning its reaction mechanism and regiocontrol had to be done, necessitating for instance the synthesis of various pentadienyl buildingblocks. Finally, an ideal compound was identified, facilitating the desired Tsuji-Trost reaction giving the target intermediat as the sole isomer. The synthesis of bakkenolid-A could not be completed, due to significant steric hindrance during the final Diels-Alder step. The second part of this work dealt with the total synthesis of macrocidin A. A fundamental part of the retrosynthetic analysis was the 3-acylation of tetramic acids using Yoshii's protocol. Since its yields tend to be unpredictable due to its still unknown mechanism, scope and limitation were to be examined in detail. A thorough screening showed the yields to be highly depending on chain length and degree of α-branching of the used carboxylic acids. The 3-acyl-tetramic acids thus obtained were subsequently deprotected either partially or globally giving access to a library of 3-acyl-tetramic acids. These were tested for cytoxic and antibiotic properties. Some compounds showed IC50 values of less than 10 µM. However, a stringent structure-activity-relationship could not be established. The next crucial step en route to a total synthesis of macrocidin A was the closure of its 17-membred macrocycle. Originally only meant for deprotection of the phenol-function in some precursors, direct ring closure of simplified test systems could be achieved. To our great delight, by this a completely new type of macrocyclization has been discovered. This is the more valuable since it shows first signs of generality. The mechanism could so far be described as a formal Williamson type etherification; although further experiments showed the presence of palladium to be crucial for ring closure. Slight modifications to the newly found macrocyclization method also facilitated the ring closure of other 3-acyltetramic acid, thus giving access to a macrocycle, beeing an impor-tant precursor of nor-macrocidin A. Beside an extremely laborious purification, that leads to unsatisfactory yields, this newly found reaction could be a milestone towards a synthesis of the macrocidins. Both, nor-macrocidin A and macrocidin A might finally be accessible using this new macroetherification strategy
