Abstract

Ziel dieser Arbeit war die Etablierung des von OSTERMEIER und SCHOBERT entwickelten Baukastenprinzips zur Synthese chimärer Makrolid-Antibiotika sowie die Optimierung der dabei als Schlüsselreaktion verwendeten, neuartigen WITTIG-Makrocyclisierung von ω-Hydroxy-Hemiacetalen auf nur eine Stufe. Ausgehend von einem Saccharid-Baustein und einer bifunktionellen Seitenkette sollte dazu zuerst das Grundgerüst ausgewählter geeigneter Naturstoffe mit Makrolid-Struktur in Form eines ω Hydroxy-Hemiacetals aufgebaut werden, bevor im Anschluss daran dann der Ringschluss zum Makrolakton mit Ketenylidentriphenylphosphoran als Cyclisierungs-Reagenz vollzogen werden konnte. Nach Totalsynthese der dazu geeigneten makrolidischen Naturstoffe sollte abschließend eine Glykosylierung dieser mit D-Desosamin, dem für die pharmakologische Wirkung essentiellen Amino-Zucker, erfolgen, um dadurch potentielle Makrolid-Antibiotika zu generieren. Weiter sollte dann eine Derivatisierung dieser durch Variation der Seitenkette erfolgen, um so den Einfluss der Ringgröße auf Makrocyclisierung und Wirkspektrum zu evaluieren. Zur Etablierung dieser Methodik wurden 14- und 18-gliedrigen Makrolaktone aufgrund ihrer charakteristischen Strukturen ausgewählt.
Während die geplante Totalsynthese von Pestalotioprolid A eingestellt werden musste, da zwischenzeitlich eine Revision dessen postulierter Struktur ohne fundierte Berichtigung erfolgte, scheiterten die Totalsynthesen von Gliomasolid C sowie den daraus abgeleiteten Naturstoffen Sch-725674 und Gliomasolid B aufgrund einer unerwarteten, im Anschluss an die WITTIG-Makrocyclisierung ablaufenden intramolekularen MICHAEL-Addition, die zur Abschnürung eines Furan-Rings im Makrocyclus führte und alle weiteren nötigen Transformationen zur Fertigstellung der Naturstoffe unterband. Jedoch konnte dann durch die Totalsynthese von (+)-Aspicilin (12 lineare Stufen, 17% Gesamtausbeute) die nun auf eine Stufe optimierte WITTIG-Makrocyclisierung als neue Methode zur Makrocyclisierung von ω Hydroxy-Hemiacetalen etabliert werden. Anschließend wurde durch die Glykosylierung des Aglycons mit D-Desosamin zum Glykosid das Baukastenprinzip zur Synthese chimärer Makrolid-Antibiotika erstmals in seiner vollen Gesamtheit vollzogen. Dies wurde dann auch 2017 im Journal of Organic Chemistry publiziert.
Im Anschluss an die erfolgreicher Totalsynthese von (+)-Aspicilin und dessen Desosamin-Glykosid sollte noch die Derivatisierung zur Abschätzung der biologischen Aktivität anhand verschiedener Ringgrößen erfolgen. Dabei sollte zusätzlich der Einfluss dieser auf die WITTIG-Makrocyclisierung evaluiert werden. Es wurden dazu analog der Synthese von (+)-Aspicilin 12-, 14-, und 18-gliedrigen ω-Hydroxy-Hemiacetale synthetisiert, so dass nach Einbau des C-2-Körpers bei der Makrocyclisierung mit Ketenylidentriphenyl¬phosphoran entsprechend 14-, 16- und 20-gliedrige Derivate resultieren sollten. Es zeigte sich, dass kleinere Ringe eher zur Ausbildung des bereits in der Synthese von Gliomasolid C und (+)-Aspicilin beobachteten Furan-Gerüstes durch eine unerwünschte intramolekulare MICHAEL-Addition neigten, als größere. Dies ist wahrscheinlich der räumlichen Nähe der 6-Hydroxy-Funktion zum α,β-ungesättigten Lakton geschuldet und konnte auch unter milden Bedingungen nicht unterdrückt werden. Bei größeren Makrocyclen konnte diese Überreaktion nicht, bzw. nur unter sehr harschen Bedingungen beobachtet werden. Aufgrund der zunehmenden Anzahl an Freiheitsgraden verläuft die neuartige WITTIG-Makrocyclisierung jedoch bei größeren Ringen zunehmend schleppend. Insgesamt wurde weder von dem Desosamin-Glykosid des (+)-Aspicilins ausreichend Material für antibiotische Aktivitätsstudien erhalten, noch konnte aufgrund der Ergebnisse der Derivatisierung von eine Glykosylierung der nur in geringsten Mengen isolierten Derivate vorgenommen werden, so dass auch hier keine Möglichkeit für weitere Untersuchungen des biologischen Potentials bestand.

The aim of this work was the establishment of the modular principle for the simple synthesis of chimeric macrolide antibiotics developed by OSTERMEIER and SCHOBERT as well as the optimization of the novel WITTIG macrocyclization of ω-hydroxy hemiacetals to one step, which plays the key role in that methodology. Starting from a sugar building block and bifunctional sidechain the backbone of selected naturally occurring macrolides should be synthesized in form of an ω-hydroxy hemiacetal. Afterwards the macrocycle can be closed by reaction of them with ketenylidenetriphenylphosphorane as the special reagent for cyclization. After the total synthesis of this natural products the deoxygenated amino sugar D-desosamine which is crucial for biologic activity should be attached by glycosylation to gain potential new antibiotic macrolides. Further the derivatization of those synthesized macrolides was scheduled to investigate the new method of macrocyclization and the range of their efficacy in dependence of ring size. Due to their characteristic structures, the macrolides pictured in were chosen for the further establishment of this methodology.
While the total synthesis of pestalotioprolide A had to be ceased because the postulated structure of it was repealed without any rectification, the total synthesis of gliomasolide C as well as that of derived compounds sch-725674 and gliomasolide B failed due to an unexpected intramolecular MICHAEL addition, which immediately takes place after the WITTIG macrocyclization. Hereby furan derivatives were formed preventing any further transformation, so the last steps of the synthesis were not fully accomplished. However, by the total synthesis of (+)-aspicilin (12 linear steps, 17% overall yield) the optimized one-step WITTIG macrocyclization was established as new method for macrocyclization of ω-hydroxy hemiacetals. Afterwards the modular principle for the synthesis of chimeric macrolide antibiotics was conducted the first time in full length by glycosylation of the aglycone by D-desosamine to furnish the potential macrolide antibiotic. This procedure was finally published in the Journal of Organic Chemistry.
After the successful total synthesis of (+)-aspicilin and its desosamine glycoside their derivatization was outstanding for evaluation of the biological activity as well as of the WITTIG macrocyclization in dependence of ring size. Therefor, 12-, 14- and 18-membered ω-hydroxy hemiacetals were synthesized in analogy to the synthesis of (+)-aspicilin. After insertion of the C-2 building block resulting from macrocyclization with ketenylidenetriphenyl¬phosphorane there should be obtained 14-, 16- and 20-membered aspicilin derivatives according to the developed procedure. Thereby it was shown, that smaller rings eagerly tend to the formation of the undesired furan derivatives through intramolecular MICHAEL addition then bigger ones, what was already observed in the planned total synthesis of gliomasolide C and that of (+)-aspicilin. This is presumably owed to the smaller distance between the 6-hydroxy group and the α,β-unsaturated lactone moiety and could not be suppressed by carrying out the reaction under milder conditions. By bigger macrocycles this reaction was not or only in traces observed under harsh conditions. But due to their increased number of degrees of freedom the WITTIG macrocyclization takes place more sluggishly. So, it was unfortunately not possible to get neither enough substance to study the biological activity of the desosamine glycoside of (+)-aspcilin nor to synthesize sufficient amounts of macrolide derivatives of for further glycosylation reactions of them.
