Directed cell motion is important for a large number of biological processes including wound healing, embryogenesis, synapse formation, and the immune response against pathogens. Cells direct their motion by physically and biochemically sensing their environment. This chapter presents recent techniques for studying cell migration in response to biochemical and thermal stimulation of single cells. The techniques are based on chemical and thermal microsources, holographic optical traps, and digital image processing. We show examples for the use of optically controlled chemical microsources to stimulate chemotaxis and to induce local perturbations of the cytoskeleton. Furthermore, we show that thermotaxis can be induced by using an optically controlled thermal microsource. The examples of optically controlled chemical and thermal microsources presented in this chapter show that light robotics are a highly useful and versatile tool for cell biological and biophysical studies.
