Abstract

Polyacrylamides are structurally more stable and more hydrophilic alternatives to polyacrylates and hold great potential in biomedical applications. In this study, we developed well-defined polyacrylamide-based cationic polymers with secondary and tertiary amine side-chains to systematically investigate structure–property-relationships for in vitro gene delivery. Polymethylacrylate precursor polymers with different degrees of polymerization (DPs) were first synthesized via Cu(0)-mediated radical polymerization and subsequently amidated to generate a polymer library that varied in DP, secondary/tertiary amine ratio, and side-chain ethyl substitution. We examined the effects of molar mass and cationic group composition of the obtained polyacrylamides on polymer-plasmid DNA complex formation, cytotoxicity, and plasmid DNA transfection efficiency, as well as antimicrobial activity in a preliminary screening. The results showed that within the investigated DP range, higher DP led to enhanced transfection efficiency, and the formed polyplexes were well tolerated under the tested conditions. In addition, the degree of ethyl substitution on the polymer side chains influenced polyplex size, particularly in the DP250 series, with higher ethyl incorporation leading to smaller polyplex sizes. However, the achieved transfection efficiency was generally lower compared to the industry benchmarks. The developed synthetic methods for preparing functional polyacrylamide libraries and gained insights in structure–property relationships for gene delivery and antimicrobial activity may contribute to future development of more efficient cationic polymers for transfection and as antimicrobial agents as well as for other applications of such polyacrylamides.
